The majority of common cold is caused by Rhinoviruses (HRV: 20-40%) and Coronaviruses (hCoV: 20%), while Respiratory Syncytial Virus (RSV-A/B) and Parainfluenza viruses (PIV1-4) are responsible for only 10% cases. Coronaviridae are positive-stranded RNA viruses with four common human pathogen strains 229E and NL63 from the group Alphacoronavirus, and OC43 and HKU1 from the group Betacoronavirus. Coronavirus is well known from the 2003 SARS-CoV pandemy in Asia with more than 800 fatal cases and the recent MERS-CoV pandemy with more than 500 fatalities alone in Saudi Arabia.
1 Vial red cap 96 reactions hCoV OC43 (lyophilized)
1 Vial black cap Positive Control (Cp ~ 30, lyophilized)
Additional Reagents required
LightCycler® Multiplex RNA Virus Master
A 100 bp long fragment from the Polyprotein gene is amplified with specific primers and detected with a R6G labeled hydrolysis probe (580 channel).
This assay detects 10 genome equivalent copies or less per reaction.
Sample Material and Extraction
Typical clinical samples are nasopharyngeal swabs, tracheal aspirates or bronchoalveolar lavage. See ModularDx Document Extraction Protocols
