X-linked hypogammaglobulinaemia (XLA) belongs to the primary immunodeficiencies (PID). Affected patients do not generate mature B cells and develop serious and even fatal infections if not treated. XLA is very rare (1:100,000 in male newborn). Detection of low or even missing Kappa-deleting Recombination Excision Circles (KRECs) is an indication for the presence of XLA.

Additional Reagents required
LightCycler® Multiplex DNA Master
or LightCycler® 480 Probes Master
An about 100 bp long fragment generated from Kappa-deleting Recombination Excision Circles is amplified with specific primers and detected with a LC610 labeled hydrolysis probe.
This assay must be combined with an extraction control, for example MSTN 66-0905-96, Cat. No. 07 225 253 001 (660 label) or 58-0905-96, Cat No. 07 559 968 001 (580 label). This assay can be combined with TREC 53-0621-96, Cat. No. 07 093 861 001(FAM label).
This assay detects 10 genome equivalent copies or less per reaction (plasmid DNA dilution).
