Infectious diarrhea can be caused by viruses, bacteria or parasites. Campylobacter are the most common cause of bacterial diarrhea, but infections by Salmonella, Clostridium, some strains of E. coli and less frequent Shigella, Yersinia, Plesiomonas, Aeromonas or Tropheryma can be also causative.
1 Vial blue cap 96 reactions Plesiomonas (lyophilized)
1 Vial black cap Positive Control (32,000 copies, lyophilized)
Additional Reagents required
LightCycler® Multiplex DNA Master
or LightCycler® 480 Probes Master
A 117 bp long fragment from the gyrB gene is amplified with specific primers and detected with a LC640 labeled hydrolysis probe.
In a clinical study including 1,281 diarrhea fecal samples the prevalence for Plesiomonas was 0.2%. Sensitivity of this assay as part of a hexaplex test was 100%, the specificity was 100%, both NPV and PPV were 100%.
Negative results can be considered as true results. Since the PPV for other bacteria was only 50% any PCR-positive results should be verified by cultural methods.
This assay detects 10 genome equivalent copies or less per reaction (plasmid DNA dilution).
Sample Material and Extraction
Typical samples are from feces, rectal swabs or bacterial culture. See ModularDx Extraction Protocols.
