Antibiotics containing a ß-lactam ring like penicillins and cephamycins are commonly used for treatment of Gram-positive as well as some Gram-negative bacteria. Beta-lactamases cleave the β-lactam ring. Carbapenemases, member of the Extended-spectrum ß-lactamases (ESBL) act also on carbapenems. Carbapenem-Resistant Enterobacteriaceae (CRE) are a growing threat for hospital acquired infections.

The New Delhi Metallo-β-Lactamase was originally described from New Delhi in 2009 and is now widespread in Escherichia coli and Klebsiella pneumoniae from India and Pakistan. NDM-1 carrying bacteria have been introduced also to other countries (including the United States and UK).

Additional Reagents required

LightCycler® Multiplex DNA Master
or LightCycler® 480 Probes Master

A 106 bp long fragment from the blaNDM-1 gene is amplified with specific primers and detected with a FAM labeled hydrolysis probe. This kit detects NDM-1 to NDM-13, NDM-16, NDM-17 (BLAST results).

In a German study including 60,000 samples only two cases were detected. Since PCR eventually reports results which can be not verified in culture only negative results should be considered as true.

This assay detects 10 genome equivalent copies or less per reaction (plasmid DNA dilution).
