Human parainfluenza viruses (four serotypes hPIV1-4) are negative strand ssRNA viruses belonging to the paramyxovirus. They cause flu-like illness; after the Respiratory Syncytial Virus (RSV) they are the second main cause of hospitalisation of young children suffering from respiratory illness. Infection of immunosuppressed people can cause severe pneumonia which can be fatal.

PIV-1 and -2 are the most important cause for croup (laryngotracheobronchitis) in children under 5 years. Biennial epidemics starting in fall are associated with both types while PIV-2 can have yearly outbreaks.
PIV-3 is closely associated with bronchiolitis and pneumonia in young children under the age of 1. PIV-4a/b is often not included in testing but are believed to be more common than previously thought, because most children by the age of 10 are sero-positive. PIV-4 causes less often severe disease.

Additional Reagents required
LightCycler® Multiplex RNA Virus Master

For PIV-1 a 114 bp long fragment from the HN gene, for PIV-2 a 120 bp long fragment from the L gene, for PIV-3 a 95 bp long fragment from the M gene, and for PIV-4 a 148 bp long fragment from the NP gene are amplified with specific primers and detected with Cyan500 labeled hydrolysis probes.

This assay detects 10 genome equivalent copies or less per reaction (in vitro transcribed RNA).

Sample Material and Extraction
