Morbus Whipple is a very rare disease caused by Tropheryma whipplei (Incidence less than 1:1000,000). The four classic signs are loss of weight, abdominal pain, diarrhoea and arthralgia. Infectious diarrhea can be caused by several viruses, bacteria or parasites.

A 75 bp long fragment from the 'genomic repeated sequence' is amplified with specific primers and detected with a FAM labeled hydrolysis probe.
The assay was compared in a clinical study of 640 diarrhea fecal samples from Africa with a reference assay based on the rpoB gene. The prevalence for T. whipplei was 37%. Sensitivity and specificity were 100%, NPV and PPV were 100%.
This assay detects 10 genome equivalent copies or less per reaction (plasmid DNA dilution). PCR has been tested to work between 52°C and 64°C annealing temperature.
