The innovative design of the thermal block also requires tailor-made reaction devices which match the exact geometry of the block and permit optimal heat transfer.

Plates are supplied in resealable, nonsterile plastic bags. Sealing Foils are supplied in a separate box (also contained in the plate pack).

Note: Before loading the PCR multiwell plate into the LightCycler® 480 Instrument, it has to be sealed properly with the self-adhesive Sealing Foil by pressing it firmly to the plate surface using the Sealing Foil Applicator provided with the instrument. When sealing the multiwell plate, apply the Sealing Foil Applicator several times using sufficient pressure, especially for those wells at the outer edge of the plate.
Sealing the plate is crucial to eliminate evaporation at high temperatures. Use only the recommended Sealing Foil.

During Roche production of the sealing foil, DNAses and RNAses are thermally destroyed.
Always centrifuge the filled and sealed plate before loading it into the instrument. The Foil should be applied at the end of the PCR setup, as repeated opening and closing will lead to cross-contamination.


Alternative Seal for the Microplates

Since the geometry of the microplates is compatible with the standard dimensions of multiwell plates, any standard seal, such as plastic PCR caps may be used to cover the plates. However, when the microplates are inserted into the LightCycler® 480 Instrument, the adhesive LightCycler® 480 Sealing Foils must be used for the following reasons:
  • Transparency of the LightCycler® 480 Sealing Foils is tested to give the best results when detecting fluorescent signals.
  • The foil is pressed onto the plate with a force of more than 500 N, thus producing a tight seal. The use of a different cover may lead to a leak as it may not fit the plate perfectly.
