Error-free PCR detection of viral RNA in human samples can be challenging because of

a) co-purification of PCR inhibiting substances from many human samples, or
b) errors in the purification or RT-qPCR processes.

In both cases, negative results cannot be clearly interpreted. Therefore, a generic positive internal control that monitors the whole process, and not only the qPCR step, is recommended in order to rule out false negative results. The Control RNA of the RNA Process Control Kit combines a number of important features, recommended for unambiguous RT-qPCR result interpretation.
The Control RNA is nuclease-resistant so that it can be added directly to the sample material.
The sequence is chosen to not interfere or compete with the viral target analysis in any of the process steps

RNase-resistant Control RNA is added directly to the sample with in a defined concentration and is used to control the complete workflow, from RNA extraction to RT-qPCR. The co-isolated Control RNA is detected in a multiplex RT-qPCR reaction together with the target viral parameter(s). Detailed protocols for use with the MagNA Pure 96 Instrument and nucleic acid isolation kits are given in the product information. The performance with a large variety of human sample materials, such as blood, plasma, serum, stool, and a number of viral targets, has been extensively evaluated.

The generic internal Control RNA of the RNA Process Control Kit is encapsulated in a nuclease-resistant coat protein of bacteriophage MS2. The synthetic Control RNA sequence does not match any mammalian sequence and does not interfere with the viral target RNA during isolation or RT-qPCR. The robust PCR Detection Mix for the RNA Control is designed to function under a variety of commonly used PCR reaction conditions in multiplex with viral targets. The multiplex qPCR is also supported by the supplied LightCycler® Multiplex RNA Virus Master.
